  • A proposal was recently made to add major and minor lineages to classify DENV, addressing the challenges posed by its genomic diversity. 

  • These lineages, based on phylogenetic thresholds, remain stable across studies. 

  • They offer a useful framework for discussing DENV diversity at regional and national levels, even when using partial genome sequences.

  • This standardized approach aids in identifying and tracking lineages of epidemiological or clinical significance.

  • The new genotype definitions successfully decrease unassigned DENV sequences

  • Major lineages, labeled with Roman alphabet letters, address regional-scale questions, while minor lineages, indicated by numbers and full stops, provide finer granularity, similar to SARS-CoV-2 pango lineages. Ex. DENV-2 genotype II is widespread across DENV-affected regions.

  • The major lineage DENV-2II_A is mainly found in the eastern hemisphere.

  • Among minor lineages, 2II_A.2.1 occurs in India and Kenya, while 2II_A.2.2 has a broader distribution in South and East Asia.

  • Detecting these lineages can provide insights into potential introductions or local occurrences.
